Semimicro method for the determination of salicylate levels in blood.

چکیده
WIDE VARIETY OF METHODS are available for the determination of salicylate in body fluids and tissues. These range from the spot test type used at the patients’ bedside to the organic solvent partition methods employed in more exact studies. In recent years, the method of Brodie et al. (1) involving extraction with ethylene chloride has gained widespread acceptance. The method commonly employs 1 or 2 ml. of plasma and can be applied over a wide range of salicylate concentrations. P. K. Smith et at. (2) adapted the method to the determination of urinary salicyluric acid and salicylates. In the course of our investigations we have had occasion to determine salicylate levels in all types of body fluids, whole blood, plasma, serum, and tissue samples. At times we have been handicapped by low salicylate levels, insufficient available volume of samples, and necessity for frequent sampling. The renewed interest in the course of blood salicylate levels at various time intervals following a dose of aspirin also posed a similar problem. The majority of investigators have determined salicylate levels 30 minutes to 1 hour after the ingestion of aspirin. We have long felt that it would be of interest to study levels that occur within the first few minutes after the ingestion of the drug. Such an experiment would involve large numbers of patients to obtain venepunctures at staggered intervals. If more frequent specimens could be taken from the same patient, additional information could be obtained concerning salicylate levels versus dosage versus time.

A Semimicro Modification of the Karr Urea Determination Method Using the Somogyi Serum Filtrate.
B ECAUSE of the pressing need for conserving both the patient’s blood and laboratory time, an attempt has been made to modify the Karr urea method (1) for semimicro amounts of Somogyi filtrate (2). Since this serum filtrate was already being used for glucose in our clinical laboratory, the saving of time and material which such a method would bring-if accurate enough for clinical purposes-was o...
متن کاملThe ultramicro determination of salicylates in biologic fluids.
A method for determining salicylates in ultramicro volumes of blood, spinal fluid, and urine has been investigated. This method combines the simplicity of one reagent for color development and deproteinization with a high degree of accuracy and precision. Apparent salicylate values in blood and spinal fluid of normal individuals are of the order of 0.7 mg./100 ml. The application of this method...
متن کاملFluorophotometric method for the estimation of salicylate in blood.
While salicylates are among the most frequently prescribed drugs, methods for their determination in blood have been neither sufficiently sensitive nor precise. Brodie, Udenfriend, and Coburn (1) have devised a procedure suitable for the clinical measurement of blood salicylate levels if large doses are given. In their method the salicylate is extracted from acidified plasma with ethylene dichl...
متن کاملThe Enhancing Effect of Sodium Glycocholate and Sodium Salicylate on Rats Gastro-intestinal Permeability to Insulin
There have been numerous efforts to formulate insulin into an oral dosage form. The major problems involved with the oral administration of insulin are acidic and enzymatic decomposition by the gastric medium, and poor absorption in the small intestine due to its macromolecular structure. This study attempted to test the enhancing ability of two absorption enhancers, sodium glycocholate (Na-G...
